Resumo: | Seletividade de alquilação na posição N1 em pirimidinas (timina e uracil) foi realizada pela aplicação de N3–benzoil timina e N3-benzoil uracil como substratos, onde o átomo N3 está temporariamente protegido, expondo o átomo N1 como sítio de alquilação. Os compostos N-3 benzoilados foram obtidos pela dupla benzoilação nas posições N1 e N3, e hidrólise básica seletiva no átomo N1. Usando as condições de Mitsunobu (iPrO2CN=NCO2iPr, Ph3P, ROH) as duas pirimidinas N3-benzoiladas foram reagidas com álcool alílico e álcool propagílico. Para evitar as dificuldades de purificação dos intermediários N3-benzoil-N1-alquilados, foi realizada hidrólise básica para remover o grupo N3-benzoil, pois isto facilita separação dos produtos finais e óxido de trifenilfosfina e iPrO2CNHNHCO2iPr. N1-Alíl e N1–propargil timina e uracil são os substratos para as transformações futuras. |
Abstract: | Selectivity of alkylation at the N1 position in pyrimidines (thymine and uracil) was achieved by the application of N3-benzoyl thymine and N3-benzoyl uracil as substrates, where the N3 atom was temporarily protected, exposing the N1 atom as the alkylation site. The N3-benzoylated compounds were obtained by double benzoylation at the N1 and N3 positions, and selective basic hydrolysis at the N1 atom. Using the Mitsunobu conditions (iPrO2CN=NCO2iPr, Ph3P, ROH) both pyrimidines were reacted with allyl and propargyl alcohol. To avoid difficulties during purification of the N1-alkyl N3- benzoylated intermediates, basic hydrolysis was realized to remove the N3-benzoyl group, since this facilitates the separation of final products from triphenylphosphine oxide and iPrO2CNHNHCO2iPr. N1-Allyl and –propargyl thymine and uracil are the substrates for future transformations. |
